چکیده

Premature Chromosome Condensation (PCC) has been shown to be a powerful cytogenetic and cytokinetic tool in understanding cell-cycle analysis(1) and also for diagnostic purposes(2,3). Although PCC is a useful cytogenetic tool, it involves somatic cell hybridizations employing a mitotic partner. Understanding of the factors responsible for chromatin condensation and their mechanism of action can throw better light on the cell cycle kinetics and also the protein repertoire of the cells in the various phases of the cell cycle. A number of proteins involved at various levels of cell cycle have been characterized and a variety of protein kinases have been separated and identified in extracts from mitotic and interphase cells. It is also understood that protein phosphorylation / dephosphorylation may be integral to the mechanism of chromosome condensation(4). The induction of PCC in cells using chemicals was well demonstrated and some of the chemical agents employed ABSTRACT

Ribonucleic Acid and Protein Synthesis in Mitotic Hela Cells

HeLa cells arrested in mitosis were obtained in large numbers, with only very slight interphase cell contamination, by employing the agitation method of Terasima and Tolmach, and Robbins and Marcus. Protein synthesis and RNA synthesis were almost completely suppressed in mitotic cells. Active polyribosomes were nearly absent in mitotic cells as compared with interphase cells treated in the same...

Mitotic Chromatin Condensation In Vitro Using Somatic Cell Extracts and Nuclei with Variable Levels of Endogenous Topoisomerase II

We report the development of a new method for producing mitotic extracts from tissue culture cells. These extracts reproducibly promote the condensation of chromatin in vitro when incubated with purified interphase nuclei. This condensation reaction is not species specific, since nuclei from chicken, human, and hamster cell lines all undergo chromatin condensation upon incubation with the extra...

Cell cycle control of higher-order chromatin assembly around naked DNA in vitro

We have developed an in vitro system in which higher-order chromatin structures are assembled around naked DNAs in a cell cycle-dependent manner. Membrane-free soluble extracts specific to interphase and mitotic states were prepared from Xenopus eggs. When high molecular weight DNA is incubated with interphase extracts, fluffy chromatin-like structures are assembled. In contrast, mitotic extrac...

ORC Is Necessary at the Interphase-to-Mitosis Transition to Recruit cdc2 Kinase and Disassemble RPA Foci

The origin-recognition complex (ORC) has an essential role in defining DNA replication origins and in chromosome segregation. Recent studies in Drosophila orc2 mutants, and in human cells depleted of ORC2, have suggested that this factor is also implicated in mitotic chromosome assembly. We asked whether ORC was required for M phase chromosome assembly independently of its function in DNA repli...
